In-vitro insights to root canal treatments: Chemo preventive potential CPP-Testing of endodontic sealers for therapeutic applications

Abstract
Objective: To explore the chemo-preventive potential (CPP) testing of endodontic sealers in root canal treatments through in-vitro analysis to evaluate its cytotoxicity via cell viability assays.
Methodology: After study approval (BMU-EC/06-2022) from respective institute, three root canal sealers; Endoseal MTA (E-MTA), Ketac-Endo (Ketac-E) and Sealapex (S-apex) were used as per manufacturer’s instructions. Human periodontal ligament stem cells (PDLSCs) were sub-cultured by in-vitro experiment and treated by sealers in pre-defined serial concentrations of 20ul, 40ul and 60ul, respectively based on MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] cell viability assay.
Result: CPP Testing of Ketac-E (IC50, 22.1µl) and S-apex (IC50, 16.5µl) showed higher cytotoxicity at lower quantity as compared to E-MTA (IC50, 44µl) on Human PDLSCs.
Conclusion: This study supports the therapeutic potential of E-MTA being better in its biocompatibility, healing, and lower cytotoxicity in endodontic applications.
